Amitriptyline is the derivative of Amitriptyline (HCl). It is of Synthetic origin and belongs to Dibenzocycloheptadiene. It belongs to Adenosine uptake inhibitor pharmacological group on the basis of mechanism of action and also classified in Psychotherapeutic Drugs and Antidepressant pharmacological group.The Molecular Weight of Amitriptyline (HCl) is 313.90. How to Take It

0.285 to 0.429 mg/ kg — 6 hourly
Initial

Adult · IM
0.714 to 2.143 mg/ kg — 24 hourly
Maintenance for 6 Months

Adult · PO
50 to 100 mg — 24 hourly
At bedtime, increased gradually to 150 mg/day.

Child · Oral
0.4 to 0.8 mg/kg — 24 hourly
Recommended for 5-12 yrs Children

Safety Alerts

Amitriptyline (HCl) is contraindicated in conditions like Glaucoma, Acute recovery phase following Myocardial infarction, Prostatic hypertrophy

Important Warnings

Amitriptyline (HCl) should be used with caution in patients with cardiac disease and pre-existing hematological disorders
It is classified as a pregnancy category C drug
No complete or well-controlled studies have been done
Use during pregnancy should be avoided unless the potential benefits outweigh the possible risks to the fetus
Abrupt discontinuation should be avoided because it could precipitate symptoms of cholinergic rebound such as nausea, vomiting or diarrhea
It should be used with caution in patients with history of alcoholism or who may use sedative medicines because the depressant effect on CNS can be potentiated
Avoid Concurrent use with Cisapride
